The description of Periploca otrebla n. sp. is presented. This species is very similar to Periplocalongipenis Landry, from the Galápagos Islands. However, both of them can be separated on the basis of the male genital characters. The larvae of P. otrebla are folivorous, and they are leaf tiers on its host: Acacia macracantha Willd. (Fabaceae). The known distribution for this new species is limited to the Azapa valley, First Región of Chile. This is the first species of Periploca genus described from Chile, and the third on from the Neotropic.Se presenta la descripción de Periploca otrebla n. sp. Esta especie tiene gran afinidad con Periplocalongipenis Landry, de las Islas Galápagos. Sin embargo, ambas pueden ser separadas sobre la base de caracteres genitales del macho. Las larvas de P. otrebla son folívoras y se comportan como pegadoras de folíolos sobre su hospedero: Acacia macracantha Willd. (Fabaceae). La distribución actualmente conocida para esta nueva especie está restringida al valle de Azapa, Primera Región, Chile. Esta es la primera especie del género Periploca descrita de Chile, y la tercera del Neotrópico.
